Methods to Measure Electrical Synapse Strength
These are the six common methods used to measure electrical synapses:
1. Voltage-Clamp Recording
Voltage-clamp recording is the most sensitive method, but it is also the most invasive. A small electrode is inserted into the cell and the voltage across the membrane is recorded.
When a signal is transmitted across an electrical synapse, the voltage of the postsynaptic neuron will change. This change in voltage can be measured using voltage-clamp recording.
2. Dye coupling
Dye coupling is a less invasive method. A fluorescent dye is injected into the presynaptic neuron and a second fluorescent dye is injected into the postsynaptic neuron.
When a signal is transmitted across an electrical synapse, the dyes will mix and the fluorescence will increase. This increase in fluorescence can be used to measure the strength of the electrical synapse.
3. Two-Photon Imaging
Two-photon imaging is a less invasive method than voltage-clamp recording, but it is also less sensitive. A laser is used to excite a fluorescent dye that is only present in the presynaptic neuron.
When a signal is transmitted across an electrical synapse, the dye will be excited in the postsynaptic neuron. This can be used to measure the spread of the signal across the synapse.
4. Calcium Imaging
Calcium imaging can be used to measure the calcium influx that occurs when a signal is transmitted across an electrical synapse.
5. Fluorescence Recovery After Photobleaching (FRAP)
Fluorescence recovery after photobleaching can be used to measure the diffusion of ions across an electrical synapse.
6. Electron microscopy
Electron microscopy can also be used to visualize the gap junctions that form electrical synapses.
